ABSTRACT

Mormodica charantia (Curcubitaceae) is a plant with great medicinal potential, also used as an alternative of mosquitoes control as demonstrated by previous studies. We evaluated the larvicidal activity of crude extracts of ethyl acetate, methanol and hexane from flowers and fruits of M. charantia against Aedes aegypti (Culicidae). Flowers and fruits were macerated in methanol, ethyl acetate and hexane. Bioassays were performed with application of the extracts at final concentrations of 1 - 200 µg/mL in the middle of the third instar larvae of A. aegypti (L3). The results showed high toxicity to ethyl acetate extracts from flowers and fruits at concentrations of 200 µg/mL and 100 µg/mL, with 97% and 87% of larvae mortality (L3), respectively. Hexane extract demonstrated low toxicity, while methanol extract exhibited 78% larval mortality. The data suggested that the ethyl acetate extracts of flowers and fruits of M. charantia can effectively contribute to larvicidal activity. In addition, purification of M. charantia extracts may lead to a promising larvicidal activity to control the A. aegypti population.

ABSTRACT

The use of bladder antimuscarinics is very common in the elderly. However, recent population-based studies that assessed the use of anticholinergics or bladder antimuscarinics showed an increased risk of dementia when these drugs were used for a prolonged period. Several of these population-based studies included patients who used solifenacin, which is a bladder antimuscarinic released in 2005 with the prospect of being a more selective antimuscarinic for M3 receptors (M3R), which could make it a safer drug when trying to avoid unwanted effects of older bladder antimuscarinics such as oxybutynin, especially with regard to changes in cognition. Since the various bladder antimuscarinics have distinct pharmacological characteristics, such as in the ability to penetrate the blood-brain barrier, in selectivity for muscarinic receptors, and in brain efflux mechanisms, their effects on the central nervous system (CNS) may vary. Solifenacin was the drug selected in this review, which aims to describe the results of several articles published in recent years reporting the effects of solifenacin on cognition or the risk of dementia development. Although preclinical studies show that solifenacin can also act on brain M1 receptors (M1R), short-term clinical studies have shown it to be safe for cognition. However, there are no long-term randomized studies that prove the safety of this drug for the CNS. Thus, until the safety of solifenacin has been established by long-term studies, it seems advisable to avoid prolonged use of this drug in elderly patients.

ABSTRACT

The objective of this study was to determine the metabolizable energy, standardized ileal amino acid digestibility (SIAAD) and digestible amino acid values of wheat meal (WM), wheat gluten meal (WGM), meat bone meal (MBM) and poultry-by-product meal (PPM) in diets for broilers. In the first experiment, the total excreta collection method was used, with 180 chicks with 14 days, distributed in a completely randomized design, with 4 feeds + reference diet with 6 replicates and 6 broilers each. In the second experiment, 180 broilers from 24 to 28 days old were used, distributed in a completely randomized design, with 4 feeds + one protein free diet with 6 replicates and 6 broilers each. The AMEn values of WM, WGM, MBM and PPM were 3052, 3174, 1556 and 3200 (kcal/kg), respectively. The average SIAAD of the essential and non-essential amino acids of WM, WGM, MBM and PPM were 74.21, 97.50, 69.28 and 83.08; 76.32, 96.42, 63.93 and 77.66, respectively. The values of the main digestible limiting amino acids were 0.18, 1.42, 0.41 and 0.94 for methionine; 0.20, 1.48, 1.73 and 2.73 for lysine; 0.23, 1.67, 0.72 and 1.89 for threonine; and 0.35, 3.22, 1.38 and 2.47 for valine, respectively for the feedstuffs in study.(AU)

ABSTRACT

Queixada (Tayassu pecari) é um mamífero neotropical, classificado como vulnerável devido à caça e à destruição de seu habitat. Em razão das dificuldades na reprodução em cativeiro, técnicas de reprodução assistida podem ser aplicadas em programas de repovoamento da espécie. Dessa forma, objetivou-se identificar o melhor protocolo de teste hiposmótico (HOST) para avaliar a integridade funcional dos espermatozoides de queixada. O sêmen de quatro machos adultos foi coletado com auxílio de eletroejaculador após contenção física e protocolo de sedação e anestesia. O sêmen foi avaliado quanto às características macro e microscópicas e diluído nas seguintes soluções hiposmóticas: água destilada (0mOsmol/L), sacarose (50, 100, 150mOsm/L) e frutose (50, 100, 150mOsm/L). Cada amostra foi incubada em duplicata, e uma sofreu fixação em solução de citrato de sódio formolizado a 4%. Duzentos espermatozoides foram avaliados por amostra e classificados em reativos ou não ao HOST. Todas as soluções testadas foram semelhantes em identificar o percentual de espermatozoides reativos, independentemente de a amostra ser ou não fixada (P>0,05). Dessa forma, pode-se usar água destilada como HOST por este apresentar resultados similares e por ser um teste mais barato.(AU)


White-lipped peccary (Tayassu pecari) is a neotropical mammal classified as vulnerable due to overhunting and habitat destruction. Due to the low reproduction success in captivity, assisted reproduction techniques can be used in re-stocking programs. Therefore, we aimed to identify the best hyposmotic swelling test (HOST) to evaluate the functional integrity of peccary sperm. Semen samples of four adult males were collected with the aid of an electroejaculator after physical contention, sedation and anesthesia protocols. The semen was evaluated for macro and microscopic characteristics and diluted in the following hyposmotic solutions: distilled water (0mOsmol/L), sucrose (50, 100, 150mOsm/L) and fructose (50, 100, 150mOsm/L). Each sample was incubated in duplicate and one was fixed in 4% formalized sodium citrate solution. Two hundred spermatozoa were evaluated from each sample and classified as reactive or not- reactive to HOST. There were similar proportions of reactive and functional sperm regardless of whether the sample was fixed or not fixed (P> 0.05). Therefore, distilled water can be used as HOST to evaluate the functional integrity of white-lipped peccary sperm because it shows similar results and it is a cheaper test.(AU)

ABSTRACT

Avaliou-se a eficiência da administração de subdoses de eCG nos acupontos Bai Hui e Hou Hai em protocolos de sincronização de estro em cabras. Na primeira etapa, 57 cabras foram distribuídas aleatoriamente em quatro tratamentos: T1- 300UI de eCG intramuscular (IM); T2- 60UI de eCG no acuponto Hou Hai; T3- 60UI de eCG no acuponto Bai Hui e T4- 60UI de eCG IM; e na segunda etapa, 28 cabras foram distribuídas aleatoriamente em três tratamentos: T1- 300UI de eCG IM; T2- 30UI de eCG no acuponto Bai Hui e T3- 30UI de eCG IM. Ao final do tratamento hormonal, as cabras foram monitoradas para detecção do estro, realização das coberturas e avaliação do comportamento reprodutivo. Os dados foram submetidos à análise de normalidade, seguida dos testes estatísticos adequados para cada variável. Na primeira etapa experimental, obteve-se maior duração de estro nas cabras do T1 (P=0,009). Na segunda etapa experimental, obteve-se maior número de animais em estros no T1 (P=0,03). As demais variáveis para ambas as etapas não sofreram influência dos tratamentos (P>0,05), demonstrando que a administração de subdoses de eCG nos acupontos Bai Hui e Hou Hai foi eficiente para sincronizar o estro.(AU)


The efficiency of administration of subdoses of eCG in the Bai Hui and Hou Hai acupoints in oestrus synchronization protocols in goats was evaluated. In the first stage, 57 goats were randomly assigned to four treatments: T1- 300UI of intramuscular eCG (IM); T2- 60UI of eCG in acupoint Hou Hai; T3- 60UI of eCG in the Bai Hui acupoint and T4- 60UI of eCG IM; and in the second stage, 28 goats were randomly assigned to three treatments: T1-300UI of eCG IM; T2-30UI of eCG in the Bai Hui acupoint and T3- 30UI of eCG IM. At the end of the hormonal treatment the goats were monitored for estrus detection, and evaluation of reproductive behavior. The data were submitted to normality analysis, followed by appropriate statistical tests for each variable. In the first experimental stage, a longer duration of estrus in the T1 goats (P= 0.009) was obtained. In the second experimental stage, a greater number of animals were obtained in estrus at T1 (P= 0.03). The other variables for both experiments were not influenced by the treatments (P> 0.05), demonstrating that administration of eCG subdoses in the Bai Hui and Hou Hai acupoints was efficient to synchronize the estrus.(AU)

Although goat dairy farms in Brazil may have a higher risk of infection by Neospora caninum than beef farms, risk factor evaluation on a representative population remains to be fully established in Brazil. Accordingly, this study aimed to establish the occurrence of anti-N. caninum antibodies and factors associated with exposure in 406 blood samples from five dairy and three beef goat farms in the state of Paraíba, northeastern Brazil. Anti-N. caninum antibodies were detected by indirect immunofluorescence assay (IFA), with samples considered positive when reacting with dilution ≥ 1:50. A total of 106/406 goats (26.11%; 95% CI: 21.96-30.72%) were seroreactive comprising 2/61 (3.28%), 10/45 (22.22%), 13/50 (26.00%), 17/51 (33.33%) to 29/46 (63.04%) in dairy farms, and from 3/54 (5.56%), 12/50 (24.00%) to 20/49 (40.82%) on the beef farms. No significant associations were found in relation to age, gender, dairy versus beef farms, occurrence of abortions or mummified fetuses, and seroreactivity to N. caninum (P>0.05). In conclusion, goat farms in the state of Paraíba showed the highest occurrence of anti-N. caninum antibodies to date in Brazil.(AU)


Embora as criações caprinas de leite no Brasil possam ter maior probabilidade de risco de infecção por Neospora caninum do que as de carne, a avaliação dos fatores de risco em uma população representativa ainda não está totalmente estabelecida no Brasil. Dessa forma, este estudo teve por objetivo estabelecer a soroprevalência de N. caninum e seus fatores associados à exposição em 406 amostras de sangue de cinco fazendas de leite e três de corte provenientes do estado da Paraíba, região Nordeste do Brasil. A detecção de anticorpos anti-N. caninum foi realizada utilizando-se a reação de imunofluorescência indireta (RIFI), com as amostras consideradas positivas na diluição ≥ 1:50. No total, 106/406 (26,11%; IC 95%: 21,96-30,72%) caprinos foram sororreagentes, variando de 2/61 (3,28%), 10/45 (22,22%), 13/50 (26,00%), 17/51 (33,33%) a 29/46 (63,04%) em fazendas de leite, e de 3/54 (5,56%), 12/50 (24,00%) a 20/49 (40,82%) em fazendas de corte. Não foram observadas associações significativas entre idade, sexo, criação de leite e carne, ocorrência de abortamentos ou fetos mumificados e sororreatividade para N. caninum (P>0,05). Em conclusão, fazendas de caprinos da Paraíba mostraram as mais altas ocorrências de anticorpos anti-N. caninum até o momento no Brasil.(AU)

ABSTRACT

The aim of this study was to diagnose dermatophytosis in pets and investigate the presence of dermatophytes in their home environment. Samples from hair coat were collected from 70 pets: 47 dogs, 19 cats, three guinea pigs and one rabbit. After mycological culture, 188 samples were collected from the household environments in 26 homes: 78 from places were of predominantly used by the tutors, 66 from places used by the animals, 44 from flooring, and 24 samples from contactees. Samples were seeded on Mycosel agar, incubated at 25°C, and the colonies were identified by their macro-and-microscopic characteristics. Dermatophytes were found in 37.1% of the samples originating from the sick animals. Microsporum canis was the most prevalent species, isolated in 12 dogs and eight cats; Trichophyton quinckeanum in three guinea pigs, Microsporum gypseum in two dogs and Trichophyton mentagrophytes in one cat. Dermatophytes were found in 69.2% of the surveyed homes; 29.5% of the places/objects predominantly used by the tutors, 42.4% mainly used by the animals, 31.8% from floors, and 50% from contactees. The meeting of dermatophytes in animals and in the household environment confirms the possibility of transmission by direct or indirect contact and their importance in public health.(AU)


O objetivo deste trabalho foi diagnosticar dermatofitose em pets e pesquisar dermatófitos em seu ambiente domiciliar. Colheram-se amostras de 70 pets, 47 cães, 19 gatos, três cobaias e um coelho. Visitaram-se 26 residências dos animais positivos em cultura micológica para a doença, colhendo-se 188 amostras do ambiente: 78 de locais de uso predominante dos tutores, 66 de uso dos animais e 44 de pisos; também foram colhidas 24 amostras de animais contactantes. As amostras clínicas foram semeadas em ágar Mycosel, incubadas a 25°C, e as colônias identificadas por suas características macro e microscópicas. Dermatófitos foram isolados em 37,1% dos animais suspeitos. Microsporum canis foi o mais frequente, sendo isolado de 12 cães e oito gatos, Trichophyton quinckeanum de três cobaias, Microsporum gypseum de dois cães e Trichophyton mentagrophytes de um gato. Foram encontrados dermatófitos em 69,2% das casas pesquisadas, isolando-se esses fungos em 29,5% dos locais/objetos de uso predominante dos tutores, 42,4% de uso predominante dos animais, 31,8% de pisos e 50% dos animais contactantes. O encontro de dermatófitos nos animais e em superfícies inanimadas nas residências confirma a possibilidade de transmissão de dermatofitose por contato direto e indireto e sua importância em saúde pública.(AU)

ABSTRACT

Os objetivos deste experimento foram identificar e associar alterações radiográficas do aparato podotoclear de equinos do Regimento de Cavalaria Alferes Tiradentes da Policia Militar do estado de Minas Gerais sem histórico e sinais clínicos de doença do osso navicular. Foi avaliado um total de 33 equinos, de ambos os sexos, com idade entre 10 e 20 anos. Os dígitos torácicos foram radiografados de forma padronizada nas projeções lateromedial (LM), dorsoproximal palmarodistal 65º (DPPD) e palmaroproximal palmarodistal (SK). A radiopacidade medular aumentada em projeção SK foi a principal alteração radiográfica detectada. Essa alteração foi associada a maior número de invaginações sinovais, a maior espessura de cortical em relação à medular em exposição SK e a maior relação corticomedular em exposição LM (P<0,05). Esses achados indicam uma predisposição da população equina para desenvolver a síndrome do osso navicular, possivelmente associada ao trauma repetitivo promovido pelo constante trabalho em piso duro.


The aims of this study were to identify and associate radiographic changes of podotoclear apparatus in horses from the Tiradentes Calvary Regiment of the Military Police of Minas Gerais State without history and clinical signs of navicular disease. 33 horses from both sexes, aged between 10 and 20 years were evaluated. The thoracic digits were radiographed in a standardized manner in lateralmedial (LM), palmaroproximal-distodorsal 65o(DPPD) and palmaroproximal-distopalmar (SK) projections. The increased medullary radiopacity in SK projection was the main radiological change detected and was associated with a higher number of synoval invaginations, increased cortical thickness in relation to medulla in SK exposure and increased corticomedullar in LM exposure (P < 0.05). These findings indicate a predisposition of this population to develop navicular syndrome, which is possibly associated with repetitive trauma promoted by constant work on hard floors.

ABSTRACT

A avaliação radiográfica do casco é essencial no diagnóstico e na avaliação da laminite em equinos, contudo poucos estudos têm sido feitos em raças nacionais. O objetivo deste trabalho foi realizar a morfometria de imagens radiográficas obtidas do dígito de éguas Mangalarga Marchador (MM), a fim de obter um perfil de medidas indicadoras de laminite nessa população e comparar grupos sem (escore corporal 5 e 6) e com (escore corporal 7 e 8) sinais de sobrepeso. Não houve diferença significativa entre os valores para as medidas dos dois grupos (P>0,05), mas verificou-se correlação positiva entre a distância entre os planos da linha coronária e do ápice do processo extensor (afundamento da falange distal) e indicadores de deposição de gordura localizada no pescoço e na base da cauda. Os achados demonstram que a morfometria radiográfica do dígito dessa população de equinos Mangalarga Marchador difere da de raças estrangeiras. Além disso, nos animais estudados, éguas com escore corporal 7 e 8 não aparentaram estarem mais sujeitas à laminite, uma vez que não houve diferença entre a morfometria de éguas com escore inferior. Entretanto, as correlações positivas entre índices de obesidade e a distância de afundamento da falange distal indicam que éguas Mangalarga Marchador também estão sujeitas à ocorrência de laminite associada à obesidade...


The radiographic evaluation of the hoof is essential in the diagnosis and follow-up of equine laminitis; however, few studies have been conducted in Brazilian breeds. The aim was to perform the morphometry of radiographic images taken from the digit of Mangalarga Marchador in order to obtain a profile of measurements indicative of laminitis in this population. We also compared measurements form mares without (body score 5 and 6) and with (body score 7 and 8) signs of overweight. There was no significant difference between measurements from the two groups (P> 0.05), but there was significant positive correlation between founder distance and signs of adiposity in the neck and in the base of the tail. Findings from this study demonstrate that radiographic morphology of the hoof from this population of Mangalarga Marchador mares differ from standard international reference values. Furthermore, in this group of mares, those with body condition grading 7 and 8 did not seem to be more prone to develop laminitis, since there was no difference when they were compared with mares showing lower grades. However, a significant correlation among founder distance and signs of adiposity in the neck and base of tail indicate that Mangalarga Marchador mares are also prone to obesity-associated laminitis...

ABSTRACT

Com o objetivo de se verificarem possíveis correlações entre a falange distal, o estojo córneo e a obesidade, 55 equinos de patrulhamento provenientes do Regimento de Cavalaria Alferes Tiradentes do Estado de Minas Gerais - RCAT tiveram seus dígitos torácicos radiografados. Posteriormente, os animais foram classificados quanto ao acúmulo de tecido adiposo regional por métodos subjetivos (escore corporal e escore de pescoço) e objetivos (índice de massa corporal, circunferência do pescoço e determinação de espessura do tecido adiposo por meio de ultrassonografia na inserção da cauda e do abdômen). Na população estudada, o índice de massa corporal evidenciou que 52,6 por cento dos equinos se encontravam acima do peso. A circunferência do pescoço a 50 por cento de seu comprimento dorsal e a medida ultrassonográfica na inserção da cauda se mostraram os melhores métodos objetivos para quantificar o acúmulo de tecido adiposo. Foram identificadas particularidades radiográficas características da população estudada que diferem de parâmetros internacionais. With the aim to verify possible correlations between the spatial relationship between the distal phalanx and the hoof and signs of obesity, 55 horses from the Cavalry Regiment Alferes Tiradentes of Minas Gerais State Police - RCAT had x-rays taken from their thoracic hoofs. After that, horses were classified based on their subjective (body condition and neck scores) and objective (body mass index, neck circumferences, ultrasound measurement of fat tissue of the tail and on abdomen) methods for adiposity. Within the population, the body mass index showed that 52.6 percent of horses were overweight. Neck circumference at 50 percent of its dorsal length and ultrasound measurement of fat thickness on the insertion of the tail were the best objective methods. We identified specific radiographic characteristics of the population that were different from international parameters. Significant correlations among signs of obesity and measurements that indicate lamintis were found, suggesting association between these conditions in this horse population...

Context: Active white spot lesions (WSLs) are a great concern to clinicians. Aims: This in vitro experiment analyzed cross-sectional microhardness (CSMH) values of occlusal artificially induced active WSLs (control groups D/A, D/B and D/C) along with experimental groups where these lesions were: Exposed to an artificial high risk cariogenic challenge (HRCC) using pH cycling; treated with a glass ionomer cement (GIC) and then exposed to artificial HRCC; or a fluoride varnish (FV) and afterwards submitted to the same artificial HRCC. Materials and Methods: Sixty unerupted human third molars were sectioned buccolingually on the occlusal surface and demineralized for 42 days. One half of each tooth was allocated to control groups (D/A, D/B, and D/C) and the other were used as test groups: A (pH cycling); B (GIC + pH cycling); and C (FV + pH cycling). CSMH test was performed for sound, demineralized, and treated specimens. Statistical Analysis Used: Different depths for CSMH values did not have a normal distribution (Kolmogrov–Smirnov test) and for that matter Wilcoxon and T Test were applied (significance level of 5%). Results: Mean depth of the lesion was 120 μm. A number of samples both in the test groups (n = 37) as in control groups (n = 47) had a lower Knoo p value (softening) or surface erosion. Comparisons between control and test groups only showed statistical difference at a depth 140 μm (P = 0.010) in control group D/A and for test group B at 20 μm (P = 0.004) and at 40 μm (P = 0.007). Conclusions: This in vitro study demonstrated that the use of GIC over an artificial active WSLs and exposed to an artificially HRCC setting tend to express some effect in increased surface KHN values.

ABSTRACT

Pneumonectomy is associated with high rates of morbimortality, with postpneumonectomy pulmonary edema being one of the leading causes. An intrinsic inflammatory process following the operation has been considered in its physiopathology. The use of corticosteroids is related to prevention of this edema, but no experimental data are available to support this hypothesis. We evaluated the effect of methylprednisolone on the remaining lungs of rats submitted to left pneumonectomy concerning edema and inflammatory markers. Forty male Wistar rats weighing 300 g underwent left pneumonectomy and were randomized to receive corticosteroids or not. Methylprednisolone at a dose of 10 mg/kg was given before the surgery. After recovery, the animals were sacrificed at 48 and 72 h, when the pO2/FiO2 ratio was determined. Right lung perivascular edema was measured by the index between perivascular and vascular area and neutrophil density by manual count. Tissue expression of vascular endothelial growth factor (VEGF) and transforming growth factor-beta (TGF-β) were evaluated by immunohistochemistry light microscopy. There was perivascular edema formation after 72 h in both groups (P = 0.0031). No difference was observed between operated animals that received corticosteroids and those that did not concerning the pO2/FiO2 ratio, neutrophil density or TGF-β expression. The tissue expression of VEGF was elevated in the animals that received methylprednisolone both 48 and 72 h after surgery (P = 0.0243). Methylprednisolone was unable to enhance gas exchange and avoid an inflammatory infiltrate and TGF-β expression also showed that the inflammatory process was not correlated with pulmonary edema formation. However, the overexpression of VEGF in this group showed that methylprednisolone is related to this elevation.

ABSTRACT

Brosimum gaudichaudii Tréc. (mama-cadela) é uma planta medicinal nativa do Cerrado, utilizada na medicina tradicional. O objetivo deste trabalho foi verificar a possibilidade de propagação de mama-cadela por meio de estacas de raízes, sob o efeito de reguladores de crescimento e de diferentes substratos. No experimento 1 foram avaliados os efeitos da aplicação de ácido indol-butírico (AIB) e ácido naftaleno-acético (ANA) na concentração de 1000 mg L-1 e três substratos (S1- areia; S2 - 75 por cento de areia + 25 por cento de substrato comercial; S3 - 50 por cento de areia + 50 por cento de substrato comercial). No experimento 2 foram avaliadas 4 doses de AIB: 0 - testemunha; 250 mg L-1 (1,3426 mM ); 500 mg L-1 (2,6853 mM) e 1000 mg L-1 (5,3706 mM); e 4 doses de ácido naftaleno-acético (ANA): 0 - testemunha; 250 mg L-1 (1,2295 mM); 500 mg L-1 (2,458 mM); e 1000 mg L-1 (4,918 mM). O delineamento experimental utilizado foi de blocos ao acaso com três repetições de seis estacas por parcela. No experimento 1, os substratos compostos por areia e areia (75 por cento) + substrato comercial (25 por cento) proporcionaram os maiores Índices de pegamento. A aplicação de AIB (1000 mg L-1) proporcionou aumentos relativos de 30,8 por cento e 51,3 por cento, no IMP quando comparada com a testemunha e a aplicação de ANA, respectivamente. No experimento 2, observou-se resposta quadrática significativa da aplicação de AIB sobre o IMP. A dose de 500 mg L-1 (2,6853 mM) promoveu maior IMP. Não houve efeito significativo das doses de ANA para os parâmetros avaliados. Não houve efeito significativo relevante de substratos ou hormônios sobre os demais parâmetros avaliados. Estes resultados evidenciam o potencial de utilização de estacas de raiz de mama-cadela para a produção de mudas clonais desta espécie.


Brosimum gaudichaudii Tréc. (mama-cadela) is a medicinal plant native to Cerrado and largely used in traditional medicine. The aim of this study was to verify the propagation of mama-cadela by means of root cutting under the effect of plant growth regulators and different substrates. In experiment 1, the effects of indole butyric acid (IBA) and naphthylacetic acid (NAA) at the concentration of 1000 mg L-1 were evaluated together with three substrates (S1 - sand; S2 - 75 percent sand + 25 percent commercial substrate; S3 - 50 percent sand + 50 percent commercial substrate). In experiment 2, 4 IBA levels were evaluated: 0 - control; 250 mg L-1 (1.3426 mM ); 500 mg L-1 (2.6853 mM) and 1000 mg L-1 (5.3706 mM), as well as 4 NAA levels: 0 - control; 250 mg L-1 (1.2295 mM); 500 mg L-1 (2.458 mM) and 1000 mg L-1 (4.918 mM). The adopted experimental design was in randomized blocks with three replicates of six cuttings per plot. In experiment 1, substrates containing sand and sand (75 percent) + commercial substrate (25 percent) promoted the highest average rooting indexes. Application of IBA (1000 mg L-1) led to increases of 30.8 percent and 51.3 percent in the average rooting indexes compared to control and NAA application, respectively. In experiment 2, there was a significant quadratic response of IBA application on the average rooting index. The level of 500 mg L-1 (2.6853 mM) promoted the highest average rooting index. There was no significant effect of NAA levels for the evaluated parameters. Similarly, there was no significant effect of substrates or hormones on the remaining parameters evaluated. These results show the potential use of mama-cadela root cuttings for the production of clonal seedlings of this species.

ABSTRACT

A erva-cidreira [Lippia alba (Mill.) N.E.Brown] é uma espécie nativa utilizada como planta medicinal em todo o Brasil. O citral é responsável pela ação sedativa da erva cidreira, sendo um dos principais componentes do óleo essencial. O objetivo deste trabalho foi avaliar o comportamento agronômico, teor e rendimento de óleo essencial e de citral em 17 acessos de L. alba da coleção da Universidade de Brasília - UnB. O experimento foi realizado no período de 19/02/2005 a 18/03/2006, em latossolo vermelho, no Distrito Federal. O delineamento experimental utilizado foi o de blocos ao acaso, com três repetições, contendo três plantas por parcela. Foram avaliados os parâmetros área foliar, comprimento da haste, produção de massa fresca (folhas e hastes), produção de massa foliar seca, teor de óleo essencial e de citral, rendimento estimado de óleo essencial e de citral por planta, além da caracterização do perfil aromático do óleo essencial. Os acessos L.41 (0,75 por cento), L.45 (0,66 por cento) e L.08 (0,62 por cento) apresentaram os maiores teores de óleo essencial. O acesso L.45 (1,32 g pl-1) apresentou o maior rendimento de óleo, seguido dos acessos L.47 (0,73 g pl-1), L.41 (0,67 g pl-1), L.34 (0,67 g pl-1) e L.33 (0,62 g pl-1). Os acessos apresentaram teores de citral variando de 51,7 por cento (L.34) a 75,1 por cento (L.09), com média de 63,6 por cento e predominância do isômero geranial (36,4 por cento) sobre o neral (27,8 por cento). O acesso L.45 (0,83 g pl-1) apresentou o maior rendimento de citral. Treze acessos apresentaram o quimiotipo citral; três o quimiotipo citral-limoneno e apenas um apresentou o quimiotipo citral-mirceno. Os resultados evidenciam o potencial de produção de citral do acesso L.45, o qual poderá ser utilizado para melhorar a qualidade da matéria prima de erva-cidreira utilizada nos programas de fitoterapia no país ou em programas de melhoramento genético para esta finalidade.


"Erva-cidreira" [Lippia alba (Mill.) N.E.Brown] is a native species used as a medicinal plant all over Brazil. Citral, a major constituent of L. alba essential oil, is responsible for its sedative activity. The aim of this study was to evaluate the agronomic behavior, content and yield of essential oil and citral in 17 accessions of L. alba from the germplasm bank of University of Brasília - UnB, Brazil. The experiment was carried out from 02/19/2005 to 03/18/2006, in red latosol, at the Federal District, Brazil. The adopted experimental design was in randomized blocks, with three replicates and three plants per plot. The following parameters were evaluated: leaf area, stem length, fresh mass (leaves and stems) production, dry leaf mass production, essential oil and citral content, estimated yield of essential oil and citral per plant, besides characterization of the essential oil aromatic profile. Accessions L41 (0.75 percent), L45 (0.66 percent) and L08 (0.62 percent) showed the highest essential oil content. Accessions L45 (1.32 g pl-1) presented the highest essential oil yield, followed by L47 (0.73 g pl-1), L41 (0.67 g pl-1), L34 (0.67 g pl-1) and L33 (0.62 g pl-1). Citral content varied from 51.7 percent (L34) to 75.1 percent (L09), with an average of 63.6 percent and predominance of the isomer geranial (36.4 percent) over neral (27.8 percent). Accession L45 (0.83 g pl-1) showed the highest citral yield. Thirteen accessions presented the chemotype citral; three, the chemotype citral-limonene, and only one had the chemotype citral-myrcene. The results evidenced the potential of accession L45 to produce citral, which can be used to improve the quality of "erva-cidreira" raw material in phytotherapy programs in the country or in breeding programs.

ABSTRACT

Pfaffia glomerata ocorre em vários estados do Brasil e países limítrofes da região Sul às margens de rios e nas orlas das matas de galerias, é espécie hidrófita e heliófita. As raízes de espécies do gênero Pfaffia são usadas na medicina popular brasileira, especialmente como tônico, afrodisíaco e no controle do diabete. O objetivo deste trabalho foi estabelecer um banco de germoplasma in vitro de Pfaffia glomerata. O experimento em delineamento inteiramente casualizado foi conduzido com seis tratamentos: 1) MS + 2 por cento de sacarose + 4 por cento de sorbitol; 2) MS/2 + 2 por cento de sacarose + 4 por cento de sorbitol; 3) MS + 2 por cento de sacarose + 4 por cento de sorbitol + 2 mg L-1 de pantotenato de cálcio; 4) MS/2 + 2 por cento de sacarose + 4 por cento de sorbitol + 2 mg L-1de pantotenato de cálcio; 5) MS + 2 por cento de sacarose + 3 por cento de manitol + 2 mg L-1de pantotenato de cálcio; 6) MS/2 + 2 por cento de sacarose + 3 por cento de manitol + 2 mg L-1de pantotenato de cálcio. Os resultados obtidos foram submetidos à análise de variância e ao teste de separação de médias de Scott Knott. Os tratamentos um, três e quatro apresentaram, significativamente, o maior número de segmentos nodais por haste, quando comparados com os tratamentos dois, cinco e seis. O tratamento dois foi o mais indicado para a conservação in vitro da espécie por ter promovido menor crescimento das plantas (altura de 3,1±1,9 cm), alto índice de sobrevivência, 100 por cento de explantes com brotação e o maior número de brotos por explante, após seis meses de cultivo. Todas as plântulas produziram raízes e não houve formação de calos, também não ocorreu hiperhidricidade nos tratamentos avaliados. As plantas aclimatizadas apresentaram 100 por cento de sobrevivência no ambiente ex vitro. A manutenção de acessos de P. glomerata no banco de germoplasma in vitro é viável tanto do ponto de vista da conservação quanto economicamente.


Pfaffia glomerata occurs in several states of Brazil and its neighboring countries in the south region at riverbanks and gallery forests. It is a hydrophyte and heliophyte species. The roots of the genus Pfaffia are used in Brazilian folk medicine especially as tonic, aphrodisiac and to control diabetes. The aim of this work was to establish an in vitro germplasm bank for Pfaffia glomerata. The experiment was carried out in completely randomized design with six treatments: 1) DM + 2 percent sucrose + 4 percent sorbitol; 2) DM/2 + 2 percent sucrose + 4 percent sorbitol; 3) DM + 2 percent sucrose + 4 percent sorbitol + 2 mg L-1 calcium pantothenate; 4) DM/2 + 2 percent sucrose + 4 percent sorbitol + 2 mg L-1 calcium pantothenate; 5) DM + 2 percent sucrose + 3 percent mannitol + 2 mg L-1 calcium pantothenate; 6) DM/2 + 2 percent sucrose + 3 percent mannitol + 2 mg L-1 calcium pantothenate. Results were subjected to analysis of variance and Scott Knott test for mean grouping. Treatments 1, 3 and 4 had a significantly larger number of nodal segments per stem, compared to Treatments 2, 5 and 6. Treatment 2 was the most appropriate for the in vitro conservation of this species since it led to the lowest growth (3.1±1.9 cm height), high survival rate, 100 percent explants with sprouting, and the largest number of sprouts per explant after six months of culture. All seedlings produced root and showed no formation of calluses or hyperhydricidity under the evaluated treatments. Acclimatized plants showed 100 percent survival in the ex vitro environment. Maintaining P. glomerata accessions in an in vitro germplasm bank is viable both economically and for conservation.

Canine visceral leishmaniasis (CVL) is recognizable by characteristic signs of disease and is highly lethal. The infection, however, may be quite inapparent in some seropositive dogs, and this has raised the polemic question as to whether or not such animals can be a source of infection for Lutzomyia longipalpis, the vector of American visceral leishmaniasis (AVL). In this study we have examined 51 dogs with acute CVL from an AVL area in Pará State, northern Brazil, and compared the parasite density, amastigotes of Leishmania (L.) infantum chagasi, in the skin, lymph node and viscera of symptomatic with that of nine asymptomatic but seropositive dogs (IFAT-IgG). Post-mortem biopsy fragments of these tissues were processed by immunohistochemistry, using a polyclonal antibody against Leishmania sp. The X² and Mann Whitney tests were used to evaluate the means of infected macrophage density (p < 0.05). There was no difference (p > 0.05) in the skin (10.7/mm² x 15.5/mm²) and lymph node (6.3/mm² x 8.3/mm²), between asymptomatic and symptomatic dogs, respectively. It was higher (p < 0.05), however, in the viscera of symptomatic (5.3/mm²) than it was in asymptomatic (1.4/mm²) dogs. These results strongly suggest that asymptomatic or symptomatic L. (L.) i. chagasi-infected dogs can serve as a source of infection, principally considering the highest (p < 0.05) parasite density from skin (10.7/mm² x 15.5/mm²), the place where the vetor L. longipalpis takes its blood meal, compared with those from lymph node (6.3/mm² x 8.3/mm²) and viscera (1.4/mm²x 5.3/mm²).


A leishmaniose visceral canina (LVC) é reconhecida pelas características clínicas da doença e é altamente letal. A infecção, entretanto, pode ser totalmente assintomática em alguns cães soropositivos, o que tem levantado questão polêmica sobre a possibilidade desses animais, serem ou não uma fonte importante da infecção para o flebotomíneo, Lutzomyia longipalpis, o principal vetor da leishmaniose visceral americana (LVA). Neste estudo foram examinados 51 cães com LVC aguda, provenientes de área endêmica de LVA no Estado do Pará, Brasil, e a carga parasitária, formas amastigotas de Leishmania (L.) infantum chagasi, na pele, linfonodo poplíteo e vísceras (fígado e baço) foi comparada com a de nove cães assintomáticos soropositivos (IFAT-IgG). Fragmentos de biópsia desses tecidos obtidos post-mortem foram processados para análise através de imunohistoquímica, usando um anticorpo policlonal contra Leishmania sp. Os testes do Qui-quadrado (X²) e Mann Whitney foram usados para avaliar as médias da densidade de macrófagos infectados (p < 0,05). Os resultados mostraram que não houve diferença (p > 0,05) na densidade de macrófagos infectados da pele (10,7/mm² x 15,5/mm²) e do linfonodo (6,3/mm² x 8,3/mm²) entre cães assintomáticos e sintomáticos. Entretanto, a densidade de macrófagos infectados da víscera de cães sintomáticos (5,3/mm²) foi maior (p < 0,05) que a de cães assintomáticos (1,4/mm²). Estes resultados sugerem, fortemente, que cães naturalmente infectados por L. (L.) i. chagasi, assintomáticos ou sintomáticos, podem servir como fonte de infecção, principalmente, considerando-se que a densidade de macrófagos infectados da pele (10,7/mm² x 15,5/mm²), local onde o flebotomíneo vetor Lu. longipalpis realiza a hematofagia, foi maior (p < 0,05) que as do linfonodo (6,3/mm² x 8.3/mm²) e vísceras (1,4/mm²x 5,3/mm²).

Este trabalho objetivou caracterizar a resistência antimicrobiana de 43 cepas de Escherichia coli isoladas do açude Santo Anastácio (Fortaleza, CE), durante os meses de janeiro a abril de 2007. As cepas foram submetidas a testes de suscetibilidade a oito antimicrobianos, tendo 67,4% delas apresentado sensibilidade a todos. Observou-se resistência à tetraciclina (25,6%), ao sulfazotrin (18,6%), ao ácido nalidixíco (9,3%) e a ciprofloxacina (4,7%). Nenhuma resistência foi observada aos betalactâmicos. Nove (21%) cepas apresentaram multirresistência, com um percentual de 78% para resistência cromossômica e 56% para resistência plasmidial. A presença de bactérias resistentes pode estar relacionada ao lançamento de antimicrobianos nas águas através de esgotos, sendo que o uso inadequado dessas águas pode comprometer a saúde da população.


The aim of this study was to determine the antimicrobial resistance of 43 Escherichia coli strains isolated from the Santo Antonio weir (Fortaleza, CE, Brazil) from January to April 2007. Susceptibility tests performed with 8 antimicrobials indicated that 67.5% of the strains were susceptible to all of them. Resistance to tetracycline (25.6%), sulfazotrim (18.6%), nalidixic acid (9.3%), and ciprofloxacin (4.7%) was observed. The strains did not show resistance to betalactamics. Nine strains (21%) were multiresistant, the resistance being chromosome (78%) and plasmid (56%) mediated. The presence of resistant bacteria may be related to antimicrobials in the sewage discharged into the weir, and the inappropriate use of this body of water may pose human health risks.

ABSTRACT

A helmintosporiose, causada pelo fungo Exserohilum turcicum, é uma das principais doenças do milho-pipoca cultivado no Brasil. Devido às características da cultura, como porte da planta, extensão da área de plantio e rentabilidade econômica, o emprego de resistência genética e controle químico têm sido as principais formas de controle da doença. O emprego de agrotóxicos na agricultura tem levado riscos à saúde humana e freqüentes danos ao meio ambiente. Assim, na busca de métodos alternativos para o controle da helmintosporiose foi avaliado o efeito fungitóxico dos extratos vegetais das plantas Achillea milefollium (mil-folhas), Cymbopogon citratus (capim-limão), Artemisia camphorata (cânfora) e Rosmarinus officinalis (alecrim) no crescimento micelial de E. turcicum, em dois meios de cultura (BDA - batata-dextrose-ágar; e LCH - lactose caseína hidrolisada). Os extratos de alecrim e cânfora foram os que apresentaram maior inibição do crescimento micelial nos dois meios de cultura, enquanto que os extratos de mil-folhas e capim limão estimularam o crescimento micelial em meio LCH.


Helminthosporiose is caused by the fungus Exserohilum turcicum and represents one of the main diseases in popcorn grown in Brazil. Due to its characteristics, such as plant size, planting area extension and economic profitability, the use of genetic resistance and chemical control has constituted the main procedure against such disease. The use of pesticides in agriculture has resulted in risks to the human health and frequent damages to the environment. Thus, the fungitoxic effect of plant extracts of Achillea millefolium (yarrow), Cymbopogon citratus (lemon grass), Artemisia camphorata (camphor) and Rosmarinus officinalis (rosemary) on the mycelial growth of E. turcicum was evaluated by using two culture media (PDA - potato dextrose agar, and LCH - lactose-casein hydrolysate) in order to set alternative methods for controlling helminthosporiose. Rosemary and camphor extracts led to higher mycelial growth inhibition in both culture media, whereas yarrow and lemon grass extracts stimulated mycelial growth in LCH medium.
